Chemistry and make-up

GHK is a three-residue peptide, glycine then histidine then lysine, first isolated by Loren Pickart in 1973 from human plasma, where its concentration falls with age [A]. The molecule matters because of the histidine in the centre. The copper(II) ion is held in a near-square-planar site built from the N-terminal amine of glycine, the imidazole nitrogen of the histidine side chain, and the deprotonated backbone amide nitrogen between glycine and histidine, with the lysine side chain and axial water contributing to stability [A]. This gives a high-affinity, physiologically exchangeable copper chelate rather than a loose salt, which is the whole point: GHK is a copper delivery and buffering molecule.

The bound copper gives the complex its characteristic blue to violet colour, which is a fast visual identity and QC cue. A colourless or green-tinged “GHK-Cu” solution suggests either that the copper is not properly complexed, that the ratio is wrong, or that degradation or oxidation has occurred. Because the active entity is a 1:1 peptide-to-copper complex, the CoA should state the copper content (often expressed as percent copper or ppm), the molar ratio, HPLC purity of the peptide, and mass spec confirmation of the peptide, not just “GHK-Cu”. Free (uncomplexed) copper is an impurity worth controlling.

What it is studied for (evidence)

  • Mechanism: GHK-Cu stimulates fibroblast production of collagen, elastin, glycosaminoglycans and proteoglycans, and modulates matrix metalloproteinases so remodelling is balanced rather than purely degradative [A] (Maquart / tripeptide-copper matrix work, reviewed in Cosmetics 2018, PMC6023977). It supports angiogenesis and nerve outgrowth in wound models, acts as an antioxidant (quenches reactive carbonyls, supports Nrf2-linked and superoxide-dismutase-type responses), and delivers copper, a cofactor for lysyl oxidase and SOD [A].
  • Established (good evidence, for a cosmetic peptide):
  • Stimulates collagen and dermal matrix synthesis in human fibroblasts and in human skin; a topical facial study reported increased collagen in about 70% of volunteers, outperforming vitamin C and retinoic acid comparators [A/D — small controlled cosmetic trial].
  • Improves parameters of photoaged skin (firmness, thickness, fine lines, clarity) in several small controlled topical trials [A].
  • Accelerates wound healing and improves healing quality in animal models and in human wound settings including diabetic and surgical (post-laser, Mohs) wounds [A]. See the 2025 tripeptide wound-healing review (PMC12595317).
  • Relative to most cosmetic peptides, GHK-Cu has an unusually deep and consistent evidence base spanning biochemistry, cell work, animal wound models and small human topical trials. It is one of the better-supported peptides in this category, though the human trials remain small and often industry-linked.
  • Emerging / thin / preclinical only:
  • Broad gene-expression modulation: Pickart, Vasquez-Soltero and Margolina (2018) reanalysed Broad Institute Connectivity Map data and reported GHK altering expression of roughly 4,192 genes by 50% or more, skewed toward DNA repair, antioxidant, anti-inflammatory and proteasome pathways [A]. This is a compelling hypothesis-generating dataset from in-vitro gene signatures, not proof of clinical anti-aging effect.
  • Hair growth (dermal papilla stimulation), lung and systemic anti-fibrotic and anti-inflammatory effects: mostly cell and animal data [A/D].
  • Marketing claims vs data: Vendors describe GHK-Cu as “resetting the genome back to health,” reversing systemic aging, detoxifying, and driving dramatic results by injection. The gene data is real but in-vitro; “resets the genome” is a marketing gloss on a CMap signature. Systemic anti-aging by injection is not supported by controlled human trials, and injectable use is off-label and unproven. The solid claims are topical, cosmetic, and about skin matrix and wound quality.
  • Human clinical status: Multiple small controlled topical cosmetic and wound studies exist, which is more than most peptides can show, but there is no large pivotal trial and no medicines approval for GHK-Cu as a drug. It lives as a cosmetic ingredient and, in the US, a compounding pharmacy input.

Handling, format and stability

Supplied either as a lyophilised blue powder (pre-formed complex) or as a tinted aqueous concentrate; reconstitution is a research-handling step only, no dosing here. Store cold and protected from light; copper complexes can catalyse oxidation, so avoid prolonged heat and strong light. The blue colour is a stability tell: fading or a shift toward green or brown suggests copper release or oxidative degradation. In cosmetic formulation GHK-Cu is sensitive to pH and to chelators and strong reducing agents (for example high-dose vitamin C) that can strip or reduce the copper, so blend compatibility matters. In the Glow and KLOW blends the copper redox activity is also why formulation and CoA discipline around the other components matters.

Regulatory and trade status

  • UK/EU cosmetic: GHK-Cu is an established cosmetic ingredient under INCI name Copper Tripeptide-1, used within EU Regulation 1223/2009; it is not listed in the prohibited Annex II or restricted Annex III, and is not a labelled fragrance allergen. Typical cosmetic use is roughly 0.05 to 2%. As a research chemical a reputable supplier should still sell strictly research use only and avoid medicinal or treatment claims [B — verify current CosIng entry].
  • US: As a cosmetic, GHK-Cu has long been marketed topically (CIR-reviewed) and is regulated by the FDA as a cosmetic ingredient, not a drug. On the compounding side, GHK-Cu was NOT one of the seven peptides reviewed at the 23-24 July 2026 PCAC meeting (those were BPC-157, KPV, TB-500, MOTS-c, Semax, Epitalon and emideltide/DSIP). Instead, in April 2026 the FDA removed GHK-Cu from the interim 503A bulks lists altogether (the non-injectable route from Category 1 and the injectable route from Category 2) after the original nominations were withdrawn, and stated it intends to consult the PCAC about GHK-Cu before the end of February 2027. So GHK-Cu is currently off the interim lists and awaiting a later PCAC review, not moving through the July 2026 cohort [B] (FDA 503A bulk substances).
  • WADA: GHK-Cu is not explicitly named on the WADA Prohibited List. It is a cosmetic-category tissue peptide rather than a hormone or growth factor, so it is not obviously caught, but sport-context users should check the current list and the S0 non-approved-substance logic [B — verify].
  • Customs / trade: Lower scrutiny than GLP-1s. Main risks are copper-content and identity mislabelling rather than legal status.

Plain summary

GHK-Cu is a small copper-carrying tripeptide (Gly-His-Lys plus a copper ion) that the body makes naturally, and it has one of the strongest evidence bases of any cosmetic peptide for boosting skin collagen, supporting wound healing, and acting as an antioxidant, though the human trials are small and mostly topical. Commercially the key is the copper: a genuine product is blue, has a stated copper content and a 1:1 peptide-to-copper ratio, and the CoA should prove both the copper and the peptide, because “GHK-Cu” that is colourless or missing its copper data is a red flag.
