What it is

Semax is a synthetic heptapeptide, an ACTH(4-10) analogue, with the sequence Met-Glu-His-Phe-Pro-Gly-Pro (MEHFPGP) and a free-base molecular weight of roughly 813.9 g/mol. It is built from the ACTH(4-7) fragment with a Pro-Gly-Pro tail added at the C-terminus. That proline-rich cap blocks aminopeptidase and carboxypeptidase cleavage, so the peptide persists far longer than the native ACTH fragment. It carries one methionine (the sulfur in the formula) and one histidine, with no cysteine and no disulfide bridge. AxYn supplies it as 2 mg and 10 mg research vials, commonly as the acetate salt. Note that N-acetyl-Semax is a distinct covalently modified molecule and is not the same substance.

Research context

Semax is studied at the molecular and receptor-signalling level. Preclinical work describes upregulation of BDNF and NGF and their receptor signalling, with downstream engagement of CREB, MAPK/ERK and PI3K/Akt pathways and modulation of monoaminergic tone.
